The sidequest becomes available after you visit Draklor Laboratory. To even begin the sidequest game, you must have already completed the Cactuar Family sidequest. There are five areas you can fish from, Downstream, Midstream, Upstream, Hidden Shoals and the Den of the River Lord. You basically just fish until you get certain items which you need to complete a puzzle, which is the Fishing Sidequest.
